The library was established in 1969 known as the Penang University Library located in Malayan Teachers College, Gelugor, Penang. The library moved to Minden (temporary building) in 1971 and occupied a new building in 1979.
The Main Library II was completed in 1994. On December 10, 2004, the Main Campus Library was named the Hamzah Sendut Library in conjunction with the name of the First Vice-Chancellor of Universiti Sains Malaysia, namely Tan Sri Hamzah Sendut.
On January 11, 2011, the additional building of the Hamzah Sendut Library, which is the research wing of the Library, began operating and was subsequently inaugurated on 3 March 2011 by the former Vice Chancellor, Professor Tan Sri Dato' Dzulkifli AbdulRazak.
